and neuronal patterning during development, is also active in the adult brain, where it contributes to neurogenesis, neuroprotection, and injury response. In Alzheimer&#8217;s disease (AD), one of the leading causes of dementia, recent studies suggest that the Shh pathway is altered and downregulated, with potential implications in neurodegenerative progression. This project aims to clarify the role of the Shh pathway in AD pathogenesis, using three-dimensional brain organoids derived from iPSC cells of patients and controls.  <\/p>\t\t\t\t\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t\t<\/div>\n\t\t<\/div>\n\t\t\t\t\t<\/div>\n\t\t<\/section>\n\t\t\t\t<section class=\"elementor-section elementor-top-section elementor-element elementor-element-cdee206 elementor-hidden-desktop elementor-hidden-tablet elementor-hidden-mobile elementor-section-boxed elementor-section-height-default elementor-section-height-default\" data-id=\"cdee206\" data-element_type=\"section\" data-e-type=\"section\">\n\t\t\t\t\t\t<div class=\"elementor-container elementor-column-gap-default\">\n\t\t\t\t\t<div class=\"elementor-column elementor-col-100 elementor-top-column elementor-element elementor-element-c8cb5a8\" data-id=\"c8cb5a8\" data-element_type=\"column\" data-e-type=\"column\">\n\t\t\t<div class=\"elementor-widget-wrap elementor-element-populated\">\n\t\t\t\t\t\t<div class=\"elementor-element elementor-element-b50d6bd elementor-widget elementor-widget-text-editor\" data-id=\"b50d6bd\" data-element_type=\"widget\" data-e-type=\"widget\" data-widget_type=\"text-editor.default\">\n\t\t\t\t<div class=\"elementor-widget-container\">\n\t\t\t\t\t\t\t\t\t<p>In the first phase, the activity of the Shh pathway during organoid development and growth will be analyzed, monitoring the expression of receptors (Patched1), co-receptors (Smoothened), and the main transcriptional effectors, with particular attention to <em>Gli1<\/em>. The transcriptional targets of the pathway (e.g., <em>CyclinD2<\/em>, <em>Olig2<\/em>) will also be analyzed in relation to the appearance of classic AD pathological markers, such as A\u03b2 and phosphorylated tau. Furthermore, through immunofluorescence and confocal microscopy, the structure of <strong>primary cilia<\/strong>, essential for Shh signal transduction and potentially altered by A\u03b2 deposition, will be studied.  <\/p><p>In a second phase, genetically modified organoids will be generated to express <em>Gli1<\/em>-GFP in a controllable manner through light stimulation (optogenetic PA-Cre), in order to selectively activate the Shh pathway downstream of ciliary damage. The timing and modalities of induction will be optimized and monitored over time through the GFP signal. <\/p><p>Finally, the effect of <em>Gli1<\/em> activation will be evaluated at the phenotypic level (A\u03b2, tau, cell viability and proliferation) and transcriptional level, using <strong>OpenST<\/strong>, a spatial transcriptomics technology developed and adapted by the host laboratory for use in organoids.
